Researchers from Canada and Singapore have discovered that the ancient viruses which entered our ancestors' genomes thousands of years ago have altered the way our cells behave; the material left by dead viruses in our cells is the answer. 1,000 copies of one particular class of retroviruses, known as the human endogenous retrovirus HERV-H, is still in our genome, and while the HERV-H retrovirus DNA is dead and cannot replicate itself, it continues to send out messages telling the embryonic stem cell how to become other cells in the body, and this is what makes the cells pluripotent."

Synthetic Chromosomes Successfully Integrated Into Brewer's Yeast 107

A fully synthetic yeast chromosome, dubbed 'synIII,' has successfully replaced chromosome 3 of multiple living yeast cells. The researchers pieced together over 250,000 nucleotide bases to accomplish this feat. Dr. Jef Boeke, the lead author of the study, says, 'not only can we make designer changes on a computer, but we can make hundreds of changes through a chromosome and we can put that chromosome into yeast and have a yeast that looks, smells and behaves like a regular yeast, but this yeast is endowed with special properties that normal yeasts don't have.' Work is underway (abstract) to synthesize the remaining 15 chromosomes."

Scientist Live-Blogs His Lab's Attempts To Generate New Type of Stem Cells 20

sciencehabit (1205606) writes "In the latest twist in the story of STAP cells, a new kind of stem cell described in two Nature papers in January, a scientist is live-blogging his latest attempt to generate the cells. The papers described how subjecting cells from newborn mice to a mildly acidic solution turned them into pluripotent stem cells, the sought-after cells that can become all the body's cell types. Kenneth Ka-Ho Lee, a stem cell researcher at the Chinese University of Hong Kong, has already tried once to make the cells, following the methods published in Nature in January. That attempt failed, which Lee documented publicly on the website ResearchGate. The lack of success mirrors other reports from scientists around the world in the weeks since the papers were published, despite a more detailed set of methods posted by some of the authors on 5 March. Today, Lee posted in the comment section of his ResearchGate review that he had set up a team of four lab members to do the experiments. They will live blog the research, and promise frequent updates."

MIT Researcher Enlists Bacteria To Assemble Nanotech Materials 36

The Register reports on an approach to nanotech that combines biological computing with micro-mechanics, embodied in the work of MIT associate professor Timothy Lu. Lu's research has resulted in the creation of tiny structures assembled using modified E. coli. "Specifically," says the article, "the MIT researchers were able to put bacteria to work producing conducting biofilms, some of which were studded with quantum dots, and arranging gold nanowires. This paves the way for the development of mass manufactured cell-based material factories, and even 'living materials' that have some of the desirable properties of bones or trees, Lu confirmed." His most radical idea, says Lu, is furniture that shapes itself to cushion the user's most-stressed areas.

Electric 'Thinking Cap' Controls Learning Speed 112

An anonymous reader writes "Vanderbilt researchers say they've shown it's possible to selectively manipulate our ability to learn by applying a mild electrical current to the brain. Using an elastic headband that secured two electrodes conducted by saline-soaked sponges to the cheek and the crown of the head, the researchers applied 20 minutes of transcranial direct current stimulation (tDCS) to each subject. Depending on the direction of the current, subjects either learned more quickly, slower, or in the case of a sham current, with no change at all. As far out as this sounds, they did a pretty good job at matching the actual appearance of the faces. This is a pretty good start on a whole new use for DNA samples. Imagine a mug shot of a rapist based only on a DNA sample."

Pine Tree Has Largest Genome Ever Sequenced 71

The tree's genome is largest yet sequenced: 22.18 billion base pairs, more than seven times longer than the human genome. The team found that 82% of the genome was made up of duplicated segments, compared with just 25% in humans. The researchers also identified genes responsible for important traits such as disease resistance, wood formation, and stress response."

Time Dilation Drug Could Let Heinous Criminals Serve 1,000 Year Sentences 914

Hugh Pickens DOT Com (2995471) writes "Like something out of the movie Inception, Rhiannon Williams reports in the Telegraph that Dr. Rebecca Roache, in charge of a team of scholars focused upon the ways futuristic technologies might transform punishment, claims the prison sentences of serious criminals could be made worse by distorting prisoners' minds into thinking time was passing more slowly. 'There are a number of psychoactive drugs that distort people's sense of time, so you could imagine developing a pill or a liquid that made someone feel like they were serving a 1,000-year sentence,' says Roache. Roache says when she began researching this topic, she was thinking a lot about Daniel Pelka, a four-year-old boy who was starved and beaten to death by his mother and stepfather.

'I had wondered whether the best way to achieve justice in cases like that was to prolong death as long as possible. Some crimes are so bad they require a really long period of punishment, and a lot of people seem to get out of that punishment by dying. And so I thought, why not make prison sentences for particularly odious criminals worse by extending their lives?' Thirty years in prison is currently the most severe punishment available in the UK legal system. 'To me, these questions about technology are interesting because they force us to rethink the truisms we currently hold about punishment. When we ask ourselves whether it's inhumane to inflict a certain technology on someone, we have to make sure it's not just the unfamiliarity that spooks us,' says Roache. 'Is it really OK to lock someone up for the best part of the only life they will ever have, or might it be more humane to tinker with their brains and set them free? When we ask that question, the goal isn't simply to imagine a bunch of futuristic punishments — the goal is to look at today's punishments through the lens of the future.'"

Overuse of Bioengineered Corn Gives Rise To Resistant Pests 259

An anonymous reader writes "Though warned by scientists that overuse of a variety of corn engineered to be toxic to corn rootworms would eventually breed rootworms with resistance to its engineered toxicity, the agricultural industry went ahead and overused the corn anyway with little EPA intervention. The corn was planted in 1996. The first reports of rootworm resistance were officially documented in 2011, though agricultural scientists weren't allowed by seed companies to study the engineered corn until 2010. Now, a recent study has clearly shown how the rootworms have successfully adapted to the engineered corn. The corn's continued over-use is predicted, given current trends, and as resistance eventually spreads to the whole rootworm population, farmers will be forced to start using pesticides once more, thus negating the economic benefits of the engineered corn. 'Rootworm resistance was expected from the outset, but the Bt seed industry, seeking to maximize short-term profits, ignored outside scientists.'"

Mice carrying a mutated copy of the gene have 10-fold fewer neurons and had other brain abnormalities, a new study (abstract) suggests. Such dramatic effects on brain size and function are unlikely in human carriers of BRCA1 mutations, the authors of the study note, but they propose the findings could shed light on the gene's role in brain evolution."

New Blood Test Offers Early Warning for Alzheimer's Onset 86

Georgetown researcher (and executive dean of Georgetown's medical school) Howard Federoff has taken a "systems" approach to diagnostics for certain chronic diseases. By comparing blood samples taken from patients who subsequently developed Alzheimer's to blood samples after the disease has manifested, Federoff has identified markers and created a blood test that is described as "90 percent accurate" (the BBC article does not delve into the ratio of false positives to false negatives) in predicting whether a currently healthy patient is likely to develop Alzheimer's in the following three years. Understandably, this raises some ethical and practical questions. What would you do differently if this test came back positive for yourself? Or for a parent? There have been some successes along the way, like genetic tests for warfarin dosage, but for the most part our gains in understanding of basic biology haven't been matched by clinical advances. Now it looks like that is at long last about to change, and it's about time.

Too many people suffer and die from too many diseases that we more or less understand, but can't effectively treat. I hated it when I worked in hands-on patient care, and I hate it now in the lab. We are, finally, getting there."

But although scientists are making progress toward artificial hamburgers, even a 2-year extension from the original deadline of 2012 wasn't enough to lure applicants for PETA's prize."

Water Filtration With a Tree Branch 205

'The most common water-borne pathogens are bacteria (e.g. Escherichia coli, Salmonella typhi, Vibrio cholerae), viruses (e.g. adenoviruses, enteroviruses, hepatitis, rotavirus), and protozoa (e.g. giardia). These pathogens cause child mortality and also contribute to malnutrition and stunted growth of children.' People have been working on engineering cheaper and cheaper filtration systems for years, but now a group of researchers has found a promising and simple solution: a tree branch. 'Approximately 3 cm^3 of sapwood can filter water at the rate of several liters per day, sufficient to meet the clean drinking water needs of one person.' 'Before experimenting with contaminated water, the group used water mixed with red ink particles ranging from 70 to 500 nanometers in size. After all the liquid passed through, the researchers sliced the sapwood in half lengthwise, and observed that much of the red dye was contained within the very top layers of the wood, while the filtrate, or filtered water, was clear. This experiment showed that sapwood is naturally able to filter out particles bigger than about 70 nanometers.' The team tested E. coli-contaminated water, and the branch was able to filter out 99 percent of the bacterial cells."

UK Government Proposes Rules To Allow 'Three-Parent Embryos' 146

The technique could allow women who are carriers of mitochondrial disease to have healthy, genetically related children. But it also transfers DNA from one egg or embryo into another, a form of genetic alteration that could be passed on to future generations. Altering the genes of human egg cells or embryos in IVF procedures is now forbidden in the United Kingdom."

3-D Printed Pelvis Holding Up After 3 Years 82

An anonymous reader writes "Here's a neat story out of Britain, with good news about long-term success for the patient involved, and for others who might benefit from similar procedures: three years ago, surgeon Craig Gerrand successfully printed and implanted an artificial pelvis (actually, about half of one) into a patient suffering from a rare form of cancer. Other techniques were ruled out, because the patient would be losing so much bone. So, after careful scanning, additive printing with titanium was used to create the replacement: 'In order to create the 3-D printed pelvis, the surgeons took scans of the man's pelvis to take exact measurements of how much 3-D printed bone needed to be produced and passed it along to Stanmore Implants. The company used the scans to create a titanium 3-D replacement, by fusing layers of titanium together and then coating it with a mineral that would allow the remaining bone cells to attach.' Now, three years after the procedure, the printed pelvis is holding up just fine, and the patient is able to walk with a cane."
